J Hepatol:在肝纤维化过程中,P2X4可调节肌成纤维细胞激活

研究背景和目的:肝纤维化以细胞外基质沉积为特征,肌成纤维细胞的激活在肝纤维化过程中,发挥着重要的作用。在肝脏受损后,细胞外ATP和它的嘌呤受体组成了强大的信号网络。虽然,肝脏高度表达嘌呤受体—P2X4 (P2RX4),但是在肝纤维化发生过程中,仍然不了解P2RX4在肝脏肌成纤维细胞 (hMF)中的作用。

研究方法:在野生型和P2x4型(P2x4-ko)基因敲除小鼠体内进行胆道结扎术,和饲喂甲氧基和胆碱缺乏的饮食。从小鼠 (野生型和 P2x4-KO基因敲除) 和人类的肝脏,分离肝脏肌成纤维细胞。采用P2X4药物抑制剂 (体外和体内)和P2X4 jsiRNAs (在体内)抑制P2X4活性。利用组织学、生物化学和细胞培养技术分析P2X4表达以及其在促纤维化和纤溶过程中发挥的作用以及在hMF激活中的作用。

研究结果:在胆管结扎或甲硫氨酸和胆碱缺乏等诱导的模型中,P2X4基因沉默或P2X4药物抑制能够降低小鼠肝纤维化程度和hMF的累积。人和小鼠hMFs表达P2X4,尤其在溶酶体中高表达。在人类和小鼠的hMFs中,P2X4的失效阻碍了hMFs激活和使得hMFs促纤维性属性减弱。研究证明P2X4调节hMF中钙进入和溶酶体胞外分泌,影响ATP释放,促纤维化因子分泌和转录因子激活。
