据诺贝尔奖官网的最新消息,瑞典斯德哥尔摩当地时间3日中午11时30分,2016年诺贝尔生理学或医学奖在瑞典卡罗林斯卡医学院颁布揭晓,日本分子细胞生物学家Yoshinori Ohsumi(大隅良典)荣获该奖项。

Yoshinori Ohsumi生于1945年,1974年在东京大学获得博士学位!然后进入Rockefeller 大学三年,1988年返回东京大学建立自己的研究室。2009年至今任东京技术研究所教授。2006年,日本学士院高度评价大隅的成就:“一贯正面致力于细胞自噬的分子机制的阐明,完全不跟风”,并授予他日本最高学术奖日本学士院奖,2012年再以极高评价获得“诺贝尔奖的最后一哩路”京都奖基础科学部门。

大隅良典教授是细胞自噬研究的先驱,曾获得京都奖、盖尔德纳国际奖及日本人第2座威利奖(2015年加拿大盖尔德纳奖揭晓)。其专长生物学,特别是分子生物学领域。最知名的成就,是阐明细胞自噬的分子机制和生理功能。由于相关论文被大量引用,因此名列2014年度汤森路透引文桂冠奖。

获奖理由:

This year's Nobel Laureate discovered and elucidated mechanisms underlying autophagy, a fundamental process for degrading and recycling cellular components.  

The word autophagy originates from the Greek words auto-, meaning "self", and phagein, meaning "to eat"Thus,autophagy denotes "self eating". This concept emerged during the 1960's, when researchers first observed that the cell could destroy its own contents by enclosing it in membranes, forming sack-like vesicles that were transported to a recycling compartment, called the lysosome, for degradation. Difficulties in studying the phenomenon meant that little was known until, in a series of brilliant experiments in the early 1990's, Yoshinori Ohsumi used baker's yeast to identify genes essential for autophagy. He then went on to elucidate the underlying mechanisms for autophagy in yeast and showed that similar sophisticated machinery is used in our cells.

Ohsumi's discoveries led to a new paradigm in our understanding of how the cell recycles its content. His discoveries opened the path to understanding the fundamental importance of autophagy in many physiological processes, such as in the adaptation to starvation or response to infection. Mutations in autophagy genes can cause disease, and the autophagic process is involved in several conditions including cancer and neurological disease

关于自噬研究背景

2015诺贝尔奖预测:细胞自噬可能上榜吗?Yoshinori Ohsumi和Noboru Mizushima是首批撰写酵母中自噬信号通路相关蛋白名单的研究人员之一。Klionski从事线粒体自噬研究,他帮助人们认识到自噬不仅仅是耗损细胞元件的一个垃圾处理系统,而是一种必不可少的细胞功能。

细胞自噬与细胞凋亡

对于上过细胞生物学这门课的同学们来说,细胞凋亡这个名词更为亲切,2002年细胞凋亡就已经获得了诺贝尔生理/医学奖,与之一脉相承的细胞自噬是否也有可能受到评审委员会的属意呢?

从概念上说,细胞自噬(Autophagy)指的是真核生物中对细胞内物质进行周转的一种进化上保守的重要过程,其实这就是存在于真核生物中一种高度保守的代谢过程,参与了调节细胞物质的合成,降解和重新利用之间的代谢平衡,影响到了生物生命过程的方方面面。

从区别上来说,细胞自噬是细胞内的一种“自食(Self-eating)”的景象,而细胞凋亡(apoptosis)是“他杀(Self-killing)”的景象,二者共用相反的安慰要素和调理蛋白,但是诱发阈值和门槛不同。

可以说细胞自噬是继细胞凋亡后,生命科学领域的又一热门研究方向,文献数量在近年来呈爆炸式增长,其中2006年以前相关文献大约就有1500条。

细胞自噬与疾病

应该说细胞自噬近年来受到热捧的一个很重要原因就在于其与疾病的关联,首当其冲是衰老。

细胞自噬的典型特征是形成自噬体并呈递给溶酶体,这一过程在蛋白质和细胞器质量控制中起基础作用并维持了细胞能量的稳态。一些研究表明,自噬与细胞衰老密切相关,参与蛋白酶和自噬相关调节的BAG蛋白家族中BAG3/BAG1比值在复制性衰老时增高,且BAG3在细胞衰老时能介导自噬的激活。研究还发现在Ras诱导的细胞衰老进程中亦可观察到较高的自噬活性。

其次是包括帕金森症在内的一些神经退行性疾病,2013年的一项研究就指出一种与细胞自噬作用相关的基因若出现异常,会导致一种罕见的脑病。这种罕见脑病被称作“伴随成人期神经退行性变性的儿童期静态脑病”(SENDA),患者大脑萎缩并伴随认知障碍

当时参与研究的科学家指出,自噬作用的异常,比如负责运送蛋白到溶酶体的自噬CMA过程出了问题,都有可能导致认知障碍。

另外一大类就是与肿瘤的关系,近年来大量的研究表明,自噬与肿瘤的发生发展密切相关,许多调节自噬的药物被用于临床肿瘤治疗的研究中。

虽然到目前为止激活细胞自噬来预防肿瘤发生的直接临床依据稍显不足,但自噬在肿瘤早期发生过程中的抑制作用已被广泛证实, 因此适当地提高基础自噬水平可能成为未来肿瘤预防的重要措施。

谁将会是哪个人?Yoshinori Ohsumi最终胜出

密歇根大学生物化学家的Daniel Klionsky曾最早在Science上发表综述介绍自噬,2005年又创办了第一本自噬杂志:《Autophagy》,同时2007年Mizushima教授也举办了第一次自噬国际会议,为自噬的宣传做了大量的工作。

东京大学的生物化学与分子生物学教授Noboru Mizushima则在2001年主要报道了Atg5的功能,这被认为是哺乳动物分子机制研究的第一环,他也参与克隆了自噬标志物LC3,制备了一些ATG基因敲除小鼠和LC3转基因小鼠,主要贡献在于自噬研究方法和与发育的关联。

Yoshinori Ohsumi则克隆了第一个酵母自噬基因Atg1和LC3,主要贡献在于酵母模型下的自噬研究。此次诺贝尔奖,Yoshinori Ohsumi最后胜出,主要在于克隆了第一个酵母自噬基因Atg1和LC3,起到决定性贡献!

此外,美国科学家Beth Levine博士首先克隆了第一个哺乳动物自噬基因Beclin,贡献也较为突出。

曾获得过诺贝尔奖的比利时科学家Christian de Duve,他在上个世纪50年代经过电镜观察到了自噬体结构,并在1963年溶酶体国际会议上首先提出了“自噬”这一说法,所以被公认为自噬研究的鼻祖。不过这位科学家已经选择了安乐死,这位一生都在研究细胞死亡方式的学者,选择了这种方式自主结束生命,不能不说冥冥之中自有安排
