使用单次培养和星形胶质细胞共培养的纯化分离的BECiBEC)测试125I -TyrA14-胰岛素摄取和转胞吞作用以及胰岛素受体(IR)封闭,胰岛素信号传导抑制,星形胶质细胞刺激和HFD的效果。

在生理胰岛素浓度下,IR而不是IGF-1受体促进BEC胰岛素摄取,这需要脂质筏介导的内吞作用,但不需要胰岛素作用于磷酸肌醇-3-激酶(PI3K)或丝裂原活化蛋白激酶激酶(MEK)。喂养HFD大鼠4周可降低iBEC胰岛素摄取并增加NF-κB结合活性,而不影响胰岛素PI3K信号,IR表达或含量或胰岛素降解酶表达。使用体外BBBiBECs和星形胶质细胞的共培养物),研究人员发现胰岛素转胞吞过程中未降解,且刺激星形细胞与L-谷氨酸增加转胞吞作用,同时抑制一氧化氮合成酶降低的胰岛素转胞吞作用。

胰岛素通过IR特异性,囊泡介导的BECs转运过程完整地穿过BBBHFD喂养,一氧化氮抑制和星形胶质细胞刺激可以调节BEC胰岛素摄取和转胞吞作用。
