然而,在常见和罕见的癌症基因和肿瘤类型中,我们对于突变的癌症基因和等位基因的序列遗传演化,以及等位基因的产生背景都不甚了解。

最近,研究人员发现,近四分之一的人类肿瘤中含有一个癌症相关基因的复合突变,定义为肿瘤中同一基因的两个或多个非同义体性突变。

复合突变富集在特定基因中,具有较高的使用率,不常见的热点突变在一定程度上受致癌适应性的驱动而获得,并以等位基因配置的方式出现,反映了背景特异性选择压力。

总的来说,复合突变是一种驱动性的改变,这种改变来自于背景和等位基因的特异性选择压力,部分取决于基因和突变功能,并导致具有生物和治疗意义的复杂性--往往是新的畸形功能。
